Which mode should I use to get as close to MLS as possible?

Which mode can I use to get as close to MLS as possible? Such as having multiple seasons, conferences, the cup, etc.

Unfortunately none, as far as I'm aware. The custom league limit is 20, so you'll have to omit three MLS teams right from the start, more if you plan on adding expansion teams like Miami and Cincinnati. I'm pretty sure there are no divisions and no playoff structure, but I could be wrong.

If you just wanted to play out season after season without worrying too much about MLS scheduling accuracy or number of teams, at least PES' standard League mode does carry over into multiple seasons. But if you really want to recreate MLS in PES as accurately as possible, your best option is probably a spreadsheet + exhibition games.
