There are many post moves that you can do to throw the defense off. The trick here is to be unpredictable. If you do the same moves over and over, the other player will adjust to it and be able to stop you. Here are a few good post dribble moves you can try:
Quick Post Spin: This move is good if you have a quick or athletic big man like Dwight Howard or Andrew Bynum. To do the quick post spin move, you need a live dribble or an unused dribble. While in the post position (Press Y if you are not in that position yet, point the left thumbstick relative to the camera to the direction where you want to quick spin to, then press Y again. For example, if you want to spin towards the baseline (if in 2K camera view), then point the left thumbstick towards the baseline and press Y. It all depends on the camera view, so play around with this move in practice mode.
Aggressive Back-down: If your post-up player is strong, or if the defender is much weaker than you, this is a good move to try out. To aggressively back-down the defender, all you need to do is press the turbo (or right trigger) while backing down the defender. This will push him harder and get you closer to the rim much faster. This move, however, is pretty risky. If the defender realizes this, they can either back off and make you fall down, or just take a charge. Use this move sparingly because this can also tire out your player faster.
Fake Post Spin: This is a great move to get the defender off-balance if you like to use the quick post spin move a lot. All the fake post spin is is a fake spin towards a certain direction to get the defender off-balance and get you more spacing to get to the basket. To perform this move, hold the left-trigger (LT) and then move the left thumbstick a certain direction to where you want to fake the spin. If you spin a certain direction a lot, you can fake spin towards that direction and then you have the other way WIDE open if they fall for it. This is very effective especially against a human opponent. Play around with this too, and remember the directions of the fake spins are camera relative as well.
