I've looked all over the internet and couldn't find an answer to this question. Can players learn signature skills overtime in association? also can they lose them?

Yes. Players will both acquire and lose signature skills over time in association mode.
Example:
Anthony Davis gained Defensive Anchor after year one
Dwayne Wade lost Finisher and Acrobat over the multiple seasons.

im wondering, what are the requirements for sig skills? because Aaron afflalo with an 88 in man def does not have LOCK DOWN, but mbah' moute with a 76 man def does. and Ilyasova, one of the top 3pt shooters last year doesn't have ANY shooting sigs
